What is Tacrolimus?
How Does It Work?
Tacrolimus binds to the cytosolic protein FKBP12 (FK-binding protein), forming a complex.
The tacrolimus-FKBP12 complex inhibits calcineurin, blocking the dephosphorylation of NFAT (nuclear factor of activated T cells).
Without NFAT dephosphorylation, cytokine gene transcription (especially IL-2) is blocked, preventing T-lymphocyte activation and proliferation that would otherwise trigger graft rejection.
