What is Mycophenolate Mofetil?
How Does It Work?
MMF is rapidly hydrolysed to the active mycophenolic acid (MPA) after oral administration, which carries out the drug's immunosuppressive action.
MPA selectively inhibits inosine monophosphate dehydrogenase (IMPDH), the rate-limiting enzyme in the de novo purine synthesis pathway required for DNA replication.
T and B cells rely almost exclusively on de novo purine synthesis. IMPDH inhibition therefore selectively suppresses lymphocyte proliferation while other cells use the salvage pathway.
