About this medicine

What is Votrient 200 mg Tablets?

Pazopanib is a targeted cancer tablet used for advanced kidney cancer (renal cell carcinoma) and for certain soft-tissue sarcomas that have spread and have already been treated with chemotherapy. It belongs to the group of medicines that block the growth of the blood vessels tumours need.

It is taken once a day, on an empty stomach, continuously. Liver function is the main thing monitored — pazopanib carries a boxed warning for liver injury — along with blood pressure, thyroid function and, at the start, an ECG. A change in hair colour (lightening or greying) is a harmless and distinctive side effect.

Pazopanib is a prescription-only medicine; a valid prescription must be provided at checkout.

How it works

How does Pazopanib work?

A tumour cannot grow beyond a few millimetres without recruiting its own blood supply. It does that by releasing VEGF, a signal that makes nearby vessels sprout towards it. Pazopanib blocks the receptors for that signal (VEGFR-1, -2 and -3) and related receptors (PDGFR, KIT) on blood-vessel and tumour cells, so the vessels do not form and the tumour is starved of oxygen and nutrients. Kidney cancer is especially dependent on VEGF signalling, which is why anti-VEGF medicines work well in it. The same receptors keep normal vessels healthy, so raised blood pressure and slower wound healing are expected effects.

Receptor blockade

Pazopanib occupies the ATP-binding pocket of VEGFR-1/2/3, PDGFR-α/β and c-KIT, switching off their signalling.

No new vessels

Without VEGF signalling, endothelial cells do not proliferate or migrate towards the tumour.

Tumour starvation

Existing tumour vessels regress and growth slows; in sarcoma and kidney cancer the direct effect on PDGFR/KIT-driven cells adds to this.

Dosage

How to take Votrient 200 mg Tablets

Your oncologist will prescribe your exact dose. The right dose for you depends on your weight, kidney and liver function, and — for some medicines — your genetic test results.

Always take exactly as prescribed. Do not change your dose or stop taking this medicine without speaking to your doctor first, even if you feel well.

Missed a dose? If it is less than 12 hours until the next dose, skip the missed one. Never take two doses together.

With food? Take on an empty stomach — at least one hour before or two hours after food. Food roughly doubles absorption. Swallow whole with water; do not crush (crushing greatly increases absorption). Avoid grapefruit.

Important safety information

Safety & warnings

Please read this section before taking Votrient 200 mg Tablets and discuss any concerns with your oncologist.

Important: Hepatotoxicity (severe liver injury)

Pazopanib can cause severe and sometimes fatal liver damage, most often in the first four months. Liver blood tests are checked before starting and on a fixed schedule during treatment; the dose is reduced or the drug stopped if they rise. Report yellowing of the skin or eyes, dark urine, pale stools, unusual tiredness, loss of appetite or pain under the right ribs immediately.

Common side effects

Side effects affect some people, not everyone. Most are manageable — let your treatment team know about anything that concerns you.

  • Diarrhoea
  • Raised blood pressure
  • Hair colour change (lightening or greying)
  • Nausea, reduced appetite, weight loss
  • Tiredness (fatigue)
  • Raised liver enzymes
  • Taste changes, mouth soreness
  • Hand-foot skin reaction, skin depigmentation
  • Underactive thyroid
  • Protein in the urine

Contact your doctor immediately if you notice:

  • Yellow skin or eyes, dark urine, pale stools, severe tiredness Stop and contact your oncologist immediately — possible liver injury (boxed warning).
  • Vomiting blood, black stools, coughing blood, or bleeding that will not stop Seek emergency care — pazopanib can cause serious bleeding.
  • Chest pain, breathlessness, sudden weakness or slurred speech Call emergency services — heart attack, stroke and blood clots have been reported.
  • Palpitations, fainting Seek urgent care — pazopanib can prolong the QT interval.
  • Severe headache, confusion, seizures, vision change Seek urgent care — very high blood pressure or a rare brain condition (PRES).
  • Severe abdominal pain with fever or vomiting Go to the emergency department — possible bowel perforation or fistula.
  • New breathlessness or dry cough Seek prompt review — lung inflammation (pneumonitis) has been reported.
  • A wound that will not heal, or planned surgery Tell your oncologist — pazopanib is stopped at least seven days before surgery.

Important: tell your doctor about these medicines

Taking Pazopanib with certain other medicines can be dangerous. Always tell every doctor, dentist, and pharmacist about all medicines you are taking.

  • Strong CYP3A4 inhibitors (ketoconazole, itraconazole, clarithromycin, ritonavir) and grapefruit Major

    Increase pazopanib levels.

  • Strong CYP3A4 inducers (rifampicin, phenytoin, carbamazepine, St John's wort) Major

Reduce pazopanib levels and may make it less effective.

  • Proton-pump inhibitors and other acid reducers (omeprazole, esomeprazole) Major

    Lower absorption of pazopanib substantially.

  • Medicines that prolong the QT interval Major

    Additive heart-rhythm risk.

Pregnancy & breastfeeding

Pazopanib can harm an unborn baby. It should not be used in pregnancy, and women who could become pregnant should use effective contraception during treatment and for at least two weeks after the last dose.

Do not breastfeed during treatment or for two weeks after the last dose.

Pazopanib may reduce fertility in men and women. Discuss fertility preservation before starting if relevant.

FAQ

Frequently asked questions

Why is my hair changing colour?

Pazopanib blocks KIT, a receptor that pigment cells in hair follicles rely on, so new hair growth can come in lighter or white. It is harmless and colour usually returns after treatment ends. Skin can lighten in patches for the same reason.

[4] Macmillan Cancer Support: Pazopanib (Votrient)

Why on an empty stomach, and why never crush the tablets?

Food roughly doubles absorption and crushing the tablet increases it much more, which would push side effects up unpredictably. Take it at least an hour before or two hours after food and swallow it whole.

How often will my liver be checked?

Because of the boxed warning, liver blood tests are taken before starting, at weeks 3, 5, 7 and 9, then at months 3 and 4, and periodically after that. Most liver-test rises are mild and settle with a dose change; the schedule exists to catch the rare serious ones early.

Can I take omeprazole with Votrient 200 mg Tablets?

Ideally not. Acid-reducing medicines cut pazopanib absorption substantially. If you need one, ask your pharmacist about timing it well away from your dose or using a short-acting antacid instead.

[1] FDA prescribing information: Votrient (pazopanib) — DailyMed

Is Votrient 200 mg Tablets chemotherapy?

No. It is a targeted therapy that blocks blood-vessel growth signals. It does not cause the low blood counts or hair loss typical of chemotherapy, though hair may change colour and tiredness is common.

[2] NCI Drug Information: Pazopanib Hydrochloride

Can I keep taking Votrient 200 mg Tablets if I need an operation?

Tell your surgeon and oncologist in advance. Pazopanib slows wound healing, so it is stopped at least a week before planned surgery and restarted only once the wound has healed.

Do I need a prescription to order Votrient 200 mg Tablets?

Yes. Pazopanib is prescription-only. Upload your oncologist's prescription at checkout; our pharmacist team reviews it before dispatch. Used as prescribed by a licensed medical professional.
