What is Ritonavir?
How Does It Work?
Even at low boosting doses, ritonavir strongly inhibits the CYP3A4 enzyme in the liver and gut wall, blocking metabolism of co-administered protease inhibitors.
Boosted PIs reach much higher plasma concentrations, allowing once-daily dosing and improving resistance barrier.
At full antiretroviral doses (600 mg twice daily) ritonavir itself inhibits HIV protease, though this is now rarely used due to tolerability concerns.
