How Does It Work?
Danazol suppresses the mid-cycle LH/FSH surge, creating an anovulatory, low-oestrogen state that causes regression of endometrial implants.
Danazol binds androgen and progesterone receptors in endometrial tissue, directly causing atrophy of ectopic endometrial deposits and relieving endometriosis pain.
In hereditary angioedema, danazol increases hepatic synthesis of C1-esterase inhibitor (C1-INH), reducing complement activation and the frequency of angioedema attacks.
How to Use
- Endometriosis: 200–800 mg/day in 2 divided doses for 3–6 months (start during menstruation)
- Hereditary angioedema: 200 mg 2–3 times daily initially; taper to lowest effective dose
- Take with food to reduce gastrointestinal side effects
