What is Tenofovir Disoproxil Fumarate?
Tenofovir DF (TDF) is a nucleotide reverse transcriptase inhibitor (NRTI) used as part of combination antiretroviral therapy (ART) to treat HIV-1 infection in adults and children. It is also approved for chronic Hepatitis B. It does not cure HIV but suppresses viral replication, reducing viral load to undetectable levels.

How Does It Work?

Cellular uptake
TDF enters HIV-infected CD4+ T cells and macrophages and is converted to its active form, tenofovir diphosphate (TFV-DP).

Chain termination
TFV-DP competes with the natural substrate dATP, incorporating into the viral DNA chain and causing premature termination of reverse transcription.

Viral suppression
Without functional reverse transcriptase, HIV cannot replicate. Consistent dosing maintains undetectable viral loads in most patients.
